Library Preparation for Medium-to High-Throughput DNA Sequencing on the Illumina Sequencing Platform. A Laboratory Protocol

Manual

This protocol achieves parallel, cost-effective preparation of Illumina sequencing libraries using only standard laboratory equipment. Costs are less than $10 per sample, and the intended scale is dozens to hundreds of samples. End-products are sequencing libraries, where each library is individually indexed with a molecular barcode. These libraries can be pooled for sequencing and subsequently demultiplexed in-silico. We provide 24 forward and 36 reverse primer sequences with a combinatorial barcoding capacity for up to 864 samples.
